Abstract
Platelet activating factor (PAF), biologically active phospholipid, is implicated on reproductive process. And PAF is found not only in mammalian spermatozoa spermatozoa but also preembryo.
This study was designed to determine the effect of PAF on spermatozoal motility, embryonal development on process of in vitro fertilization and cell culture.
In the control group which is not treated with PAF the rates of spermatozoal motility were 78¡¾12%, 62¡¾8% respectively in 4 hours and 24 hours incubations of sperm. And the rate of motility was significantly decreased by 24 hours incubation
compared
with 4 hours incubation. And in the effect of PAF according to incubation times at 4 hours and 24 hours concentrations in 10E-5M and 10E-7M, there was no significant changes of spermatozoal motility.
The fertilization rates of oocytes insemination in control group were 32.37%, 26.8% while 53.73% and 31.48% in PAF treated group respectively in 4 hours and 24 hours incubation of sperm. And the PAF treated group showed significantly an increased
rates
in fertilization compared with that in control group. But there was no differences of cleavage rates, when the fertilized ova were continuously cultured both 10E-5M and 10E-7 Min concentrations on PAF.
In conclusion, these results indicate that the treatment of PAF in process of reproduction is effective on spematozoal motility and in vitro fertilization, but there is no effectiveness on cell cleavage rates when the fertilized ova were cultured
continuously in PAF treated media.
